I just watched a talk by Rome Kanda (found out about it on Presentation Zen). The last two minutes are where he talks about the sprit of the Samurai, how a purpose or calling can become more important than life, and I liked the following thought:
Do not stop dreaming. Do not give up on your dream, trust yourself. Next time you hit a wall or are on your knees, next time you are going to give up, remember the samurai spirit…
